"tell your doctor if you are breast-feeding. You may use medroxyprogesterone injection while you are breast-feeding as long as your baby is 6 weeks old when you receive your first injection. Some medroxyprogesterone may be passed to your baby in your breast milk but this has not been shown to be harmful. Studies of babies who were breast-fed while their mothers were using medroxyprogesterone injection showed that the babies were not harmed by the medication." (emphasis added)

I have heard that the best way to make sure that Depo won't impact a mother's milk supply is to wait the 6 weeks, then she takes the progesterone-only pill for one month.  If that has no negative effect on her milk supply, then Depo should also be fine.  You can stop taking a pill, but you can't stop the shot once it's in.  
 
Just worked with a mom recently whose dr had put in a Mirena right after her baby was born.  She started with a good supply, but it tanked at 2 weeks.  The doctor wouldn't believe it was the Mirena, wouldn't even experiment with taking it out.  The Mirena website says that in rare cases the Mirena can reduce milk production.  Mom had nothing left by 6 weeks.
