You rephrased the question, but it still reads the same to me.

The drones produced by your open mated daughter queens, represent the
genetics of the mated II queen, not the drone line that the II queen was
inseminated with.

The drone line that was used to inseminate the II queen is no longer
directly accessible, without re-synthesising it using a very large
controlled breeding and selection program.
