Hobby beekeepers have perfected a fool-proof method for killing colonies, inadvertently discovered and repeatedly proven when moving colonies.  Simply close the colony up entirely.  Colonies die quickly, first put to sleep, then killed by their own carbon dioxide. 

I imagine this method would also kill the mites although it might take longer and need to be  verified.  It definitely kills bees fast.

As for how, a plastic bag is all it takes -- assuming plastic bags are still legal.
