I would suggest:
Queenless two story hive. Frames, no brood in bottom box. Queen excluder. Top box has a few frames of brood, put queens in the middle of the brood. Add bees and brood periodically.
Queen excluder cages are better, you can make them. With wire screen cages, the bees will chew the pads on the queens feet.
No more than 24 queens per hive, probably the optimum would be 12. (some will bank many more than this, but it's risky)
