One question, before I give my answer. What is the family going to do with the rest of the collection? If there are going to sell it or keep it (to sell later potentially), I think it makes more sense for the museum or association that will receive the collection to cull it themselves. I think that your criteria are smack on for determining significance for research purposes. I personally wouldn't feel comfortable about the family doing the "sampling" because I would think I might have made another decision. Of course, what I decide may not jibe with some other professionals opinion, but. . .
Also, if there are any research notes, photos, reports that go with the collection I'd get copies at the very least!

Subject: Representative artifact sample size for museum collections
Hi Everyone!
I have been contacted by a museum person who is dealing with a family that
wants to donate a representative sample of beads from a collector's estate
to a reputable museum and they are wanting to know what would be a"good
sample" size; i.e. a sample of sufficient size to allow adequate study re:
manufacturing type, shape and size range, etc.? I have my thoughts but I
want to hear what fellow HistArchers think. This information is needed to
be able to explain to the family why x number of specimens are required and
should be donated.
I would appreciate your thoughts.
