i have no idea if bees in an observation hive act differently if it's dark or light inside the hive. i've never used a propolis trap, but every description i've read or heard has sdvocated letting light in to stimulate the bees to deposit propolis...if this is true, than some behaviors are certainly affected by light.
an observation hive is wonderful for many things, but studying the interior of the broodnest, how bees move from comb to comb, or how they thermoregulate a colony with parallel combs are certainly important things to study that a single frame wide observation hive can shed very little "light" upon :)
