The 5 ml per bee space is fine but I've never seen an explanation for why there should be a max. limit for a colony. I apply 5 ml per full bee space regardless of how many frames of bees there are in the entire colony. My five frame nucs that hold my extra queens get the same treatment and they are quite strong in the spring.
