I work in a NICU and have worked in several NICU's as a lactation consultant, and I once I saw about 30 ml that a mother had pumped that was straw colored liquid that we decided was serum. 
 She had very flat nipples, and very engorged breasts, and the flanges were not placed on her nipples, and she pumped the serum. 

To help with the mother you described, I know of no test to determine if it is colostrum or not, but I would look at the mother's nipples, or watch her pumping technique. 

Maybe others have the answer to such a test.
