There is some info on my web page about Domperidone :

For the purpose of increasing supply, I don't think  it matters about the
timing of it to food.

MMM states that the dosing for increasing supply is 10-20 mg orally 3 - 4
times per day.
Concentrations reported in milk vary according to doe but following a 10 mg
dose three times daily, the avg conc in milk was only 2.6 ug/L
